START: Standardization of Single-Cell and Single-Nucleus RNA-seq Protocols for Tumors
Focus
Tissue Handling and Processing
Project Goal
To characterize human tumors with single-cell genomics in multiple centers and across platforms.
Results & Resources
This project determined the best practices for tissue processing and single-cell and single-nucleus profiling across multiple tumor types. Specifically, they compared tissue preparation and preservation, sample handling, and cell dissociation approaches; assessed experimental replicability, and performed systematic comparisons of sequencing technologies, protocols, and resulting datasets. They developed a systematic toolbox for profiling fresh and frozen clinical tumor samples, as well as openly available protocols and single-cell glioblastoma data.
