Project
Mapping the Transcriptome and Proteome of Human Testis in 3D
Focus
Reproductive
Method
Single Cell Transcriptomics, Spatial Genomics, Spatial Proteomics
Project Summary
This network will map the human testis in 3D using paired single cell analysis by mass-spectrometry, clampFISH, and RNA-sequencing.
Results & Resources
Building off previous work, this group improved SCoPE-MS, a tool to investigate the proteome of single cells, with the ability to quantify over 1,000 proteins per cell. To generate this data, they developed nPOP, a droplet sample preparation method for single-cell proteomics. With this methodology, they collected proteomic data from approximately 2,000 testis cells. To investigate the spatial transcriptome, this group developed clampFISH protocol, a high-throughput technique that, when combined with SCoPE-MS, provides insight into the transcriptome and proteome.
