Project Goal

To leverage a new multiplexed single-cell RNA-seq strategy to evaluate the effects of technical and natural variation on the single-cell profiles of peripheral blood mononuclear cells.


Results & Resources

The Ye lab developed and fine tuned both an experimental protocol (mux-seq) and computational method (demuxlet) that enables sample multiplexing for cost-effective systematic single-cell RNA sequencing. They used the method to test different methods for cryopreservation and processing, generated a mux-seq dataset of peripheral immune cells from a cohort of healthy individuals, and looked at the genetic contribution of single-cell traits.
