Project Goal
To develop and apply computational tools that utilize somatic mutations at microsatellites as genetic barcodes to reconstruct cell lineage patterns from single-cell sequencing data.
Results & Resources
The Park lab developed a computational tool, MSIprofiler, to account for the uneven allelic and genomic coverage profiles that arise due to the unavoidable genome amplification steps required to perform single-cell experiments. They have successfully used this approach to call single-nucleotide variants in single-cell whole-genome sequencing datasets.
