Project Goal
To develop psychrophilic protease methods for tissue dissociation that better preserve in vivo gene expression profiles for single-cell RNA-seq.
Results & Resources
The Potter lab developed a variety of protocols to help inform the use of cold sensitive proteases. They have shown that proteases adapted from cold sensitive organisms can be used to carry out tissue dissociation on ice, and tailored methods are necessary for each tissue. To this end, they have posted a library of protocols on protocols.io.
