Development of Site-Specific DNA-barcoded Antibodies for One-Pot Proteomics, Transcriptomics, and Interactomics
Focus
Technology Development
Project Goal
To develop high-sensitivity, DNA-barcoded recombinant antibodies to enable simultaneous quantitative ProteinSeq and RNA-Seq analysis in bulk or single-cells in a one-pot next-generation sequencing (NGS) experiment, and develop methods to interrogate cell surface interactomics by multiplexed Proximity Extension Assays.
Results & Resources
The Wells lab performed proof of principle experiments and finalized the design for ProteinSeq. The Wells lab plans to continue improving the bioconjugation chemistry in addition to improving the design and experimental protocols for the detection of protein interactions, as described in this preprint.
