Combining Light and Sound for Deep Fluorescence Microscopy

Project Summary

This team will use a novel, multidisciplinary approach aimed at overcoming the current limitations of wavefront shaping microscopy to enable truly deep biological imaging. They will combine recently developed approaches for fluorescence demixing and imaging in scattering media with hybrid imaging hardware and molecular tools. This unique approach will enable diffraction-limited imaging at several millimeters depth, by characterizing the sample’s scattering properties across spatial scales with acoustic and optical methods.

To translate this new wavefront shaping concept to practical imaging applications, they will develop and optimize novel, hybrid opto-acoustic imaging hardware as well as switchable probes that provide simultaneous, yet tunable, photo-acoustic and fluorescence contrast. This multidisciplinary approach represents a new paradigm in ultra-deep bioimaging.
